ABSTRACT

Aim To explore the effects of DADS in the cell cycle of HT-29 cells. Methods HT-29 cells growth inhibition were measured by MTT assay and cell counting. Phase distribution of cell cycle was analyzed by flow cytometry. Expression of p21~(Waf1),C-myc,Cyclin E was determined by immunohisto- chemistry analysis. Results MTT assay showed that DADS significantly inhibited HT-29 cells and exhibited a time, dose- dependent model. Adding 60 ?mol?L~(-1) and 120 ?mol?L~(-1) DADS for 24 h suppressed HT-29 growth by 23.1%,45.6% respectively. Cell counting showed that average doubling time retarded from 22.58 h in normal cultured HT-29 cells to 31.2 h in 60umol?L~(-1) DADS experimented HT-29 cells(P

ABSTRACT

Objective To investigate the clinical significance of P27~ KIP1 and S-phase kinase-associated protein 2 (SKP2) expression in human gastric carcinoma. Methods The expression of P27~ KIP1 and SKP2 was determined by SP immunohistochemical method in 69 specimens of gastric carcinoma. Results The positive rates of P27~ KIP1 and SKP2 expression were 46.38% and 33.33%, respectively. The positive rate of P27~ KIP1 expression in gastric carcinoma decreased with the poor differentiation,deep invasion and progression of pathological grade (P0.05). ABSTRACT

Objective To explore the value of morphometric quantitative parameters in differential diagnosis of common thyroid benign and malignant tumors, and establish a data bank as a basis for diagnosing diseases with the help of computer. Methods Using PIPS-2020 image analysis system to make a morphometric analysis in 32 cases of thyroid benign papillary lesion (TBPL), 32 cases of thyroid papillary adenocarcinoma(TPAC), 27 cases of thyroid follicular adenoma(TFA) and 24 cases of thyroid follicular adenocarcinoma(TFAC). Results Among many morphometric quantitative parameters of nuclei, eight including area, circumferential length, diameter,volume,long diameter, short diameter, largest to smallest diameter ratio, and circular rate were selected to analyze. The eight morphometric quantitative parameters between TFA and TFAC had a highly significant difference(P0.01). Conclusion These nuclear parameters have some value in differentiation of common thyroid benign and malignant tumors.

ABSTRACT

Aim To observe effect of DADS on the differentiation of gastric adenocarcinoma cells and acetylation of human gastric cancer tumor cell transplantable histone.Methods Human gastric adenocarcinoma heterotransplantable tumor model was constructed through subcutaneously injecting MGC803 cells to nude mice.Morphologic changes of xenograft tumor cells were observed with optical microscope,the influence of DADS on xenograft tumor cells generationcycle distribution,the expression of p21~(WAF1) protein,histone H3 and and H4 acetylion were analyzed with flow cytometry and Western blot.Results There was obvious growth inhibitory effect of xenograft tumor while abdominal injection dose were 100 and 200 mg?kg~(-1)DADS;cells density and heteromorphism decreased after treated with DADS.Flow cytometry analysis revealed that treating xenograft tumor cells with increasing quantities of DADS increased the percentage of cells in the G_2/M phase.The proportion of xenograft tumor cells in the G_2/M phase after treatment with 100 mg?kg~(-1) and 200 mg?kg~(-1) DADS was 2.22 and 3.37 times of that in NS group.Western blot analysis showed H3 acetylion increase along with G_2/M arrest of xenograft tumor cells by DADS.DADS didn′t influence the expression level of H4 acetylion;the expression of p21~(WAF1) protein in xenograft tumor increased along with the increases in the concentration of DADS.Conclusion DADS cansignificantly inhibit the growth of human gastric carcinoma xenograft in BALB/C nucle mice and induce cell differentiation,which might be related with up-regulation of histone a cetylization and p21~(WAF1) protein level.
